3' Position Modifications

The 3' position is special because it is the starting point of the synthesis. Modifications at the 3' end of the oligonucleotide are made by initiating the synthesis of the oligonucleotide on the correspondingly modified synthesis support.

The synthesis support can also be a 3' matrix for oligonucleotide affinity support (OAS) for production of oligonucleotides 3' immobilized on a matrix.

The smallest synthesis scale for oligonucleotides with 3' modifications is 0.2 μmol.
